克敏通窍止嚏汤对变应性鼻炎大鼠IgE、IL-4和IL-5含量的影响 被引量:1

摘要 目的:观察自拟克敏通窍止嚏汤对变应性鼻炎(AR)大鼠血清中IgE、IL-4和IL-5含量影响。方法:40只SD大鼠按体重随机分为正常对照组、治疗组和地塞米松(DXM)干预组,每组10只。除正常对照组外,余三组用卵清蛋白(OVA)建立AR大鼠模型,DXM干预组和治疗组分别给予DXM、自拟通窍止嚏汤中药汤剂干预14天,正常对照组用生理盐水代替OVA及干预药物。用被动皮肤过敏原试验(PCA)检验大鼠模型制备及药物干预后情况,ELISA法测定各组大鼠外周血清IL-4、IL-5和IgE。结果:模型组大鼠鼻喷嚏平均次数明显多于正常对照组、治疗组和DXM干预组(P<0.05)。模型组大鼠PCA试验阳性率为92%,其余三组全部表现为阴性。模型组IgE均显著高于正常对照组(P均<0.05);治疗组IgE、IL-4、IL-5均显著低于模型组(P<0.05)。治疗组各项指标与干预组比较差异无统计学意义(P>0.05)。结论:自拟通窍止嚏汤可降低AR大鼠IL-4、IL-5和IgE含量,减轻鼻黏膜变应性炎症。 Objective:To observe the effects of Zi-ni-ke-min-tong-qiao-zhi-ti decoction on serum levels of immunoglobin E (IgE) ,interleukin( IL)-4, and IL-5 in allergic rhinitis (AR) rats. Methods: Forty rats were equally randomized into 4 groups: normal control group,AR group,Zi-ni-ke-min-tong-qiao-zhi-ti decoction group,and dexamethasone group according to their weight. AR model was established by ovalburnin challenge method. Dexamethasone and Zi-ni-ke-min-tong-qiao-zhiti decoctiot, were administered to AR rats for 14 days. Saline was used in the nornlal control group. Passive cutaneous anaphylaxis(PCA) test was performed in all rats and tile serum levels of IgE, IL-4, and IL-5 were determined by the ELISA method. Results:The sneezing times in the AR group was significantly more than that in other 3 groups( P 〈 0.05 ). The positive rate of PCA test was 92% in the AR group and was negative in other 3 groups. The serum levels of IgE,IL-4,and IL-5 were higher in the AR group than those in the normal control group ( P 〈 0.05 ) ; those in the Zi-ni-ke-min-tong-qiao- zhi-ti decoction and dexamethasone groups were lower than those in the AR group ( P 〈 0.05 ) ; but no significant difference was noted between the dexamethasone and Zi-ni-ke-min-tong-qiao-zhi-ti decoction groups( P 〉 0.05 ). Conclusion:Zi-ni- ke-min-tong-qiao-zhi-ti decoction may reduce the serum levels of lgE, IL-4, and IL-5 to alleviate inflammation in nasal mucous membrane in AR rats.
