摘要
[目的]研究不同外植体、培养基、基因型及放置方式对蚕豆不定芽诱导效果和植株再生的影响。[方法]以蚕豆幼胚、未成熟子叶、成熟子叶、成熟子叶节为材料,通过诱导不定芽→分成单个小植株→诱导生根这一过程检测不同外植体、培养基、基因型、放置方式对不定芽诱导及植株再生的影响。[结果]所试4种外植体中,以成熟子叶节芽诱导频率最高,可达95%;不同外植体所需要的适宜培养基不同;不同基因型对蚕豆子叶节诱导芽效果差异很大;子叶节垂直半埋于培养基比平置于培养基时,其芽诱导效率高,且不定芽在IBA浓度为120mg/L的MS培养基上诱导生根,长成完整小植株。[结论]该研究建立了蚕豆器官发生再生植株的试验程序,为进一步研究提供试验依据。
[Objective] The effect of the different explants,media,genotypes and placement methods on the induction of adventitious bud and the plantlet regeneration of faba bean was studied.[Method] The immature embryo,immature cotyledon,mature cotyledon,mature cotyledon node as the experimental material,the effect of the different explants,media,genotypes and placement methods on the induction of adventitious bud and the plantlet regeneration of broad bean was studied in the regeneration system of the introduction of adventitious buds,the formation of individual small plant and its rooting.[Results] The results show that in the trial of four kinds of explants,the induction frequency of mature cotyledon culture was the highest,up to 95.0%.The medium suitable for the different explants culture was different.There was a big difference in the induction frequency from cotyledon culture to various genotypes of faba bean.The efficiency of the cotyledon vertically half-buried in the medium in its culture was better than that of it placed on the medium,which could produce higher induction rate of adventitious bud,and the adventitious buds could grow into a complete small plantlet with rooting system in the MS medium containing IBA with the concentration of 120 mg/L.[Conclusion] The process of organogenesis regeneration of faba bean was established through the experiment,which could provide a experimental basis for its further study.
