The morphology of apoptotic immune cells induced by superantigen in mice

摘要 目的:探讨超抗原(SEA)对小鼠免疫细胞的影响。方法:采用免疫学及光镜和电镜等方法观察SEA注射小鼠体内引起免疫细胞的改变。结果:SEA在小鼠体内早期引起中枢及外周免疫器官细胞增殖,表现为3H-TdR掺入量增多和淋巴细胞母细胞化;晚期则致免疫细胞凋亡,在光镜和电镜下可见细胞呈核固缩、核碎裂,其DNA琼脂糖凝胶电泳呈“梯状”改变;作为刺激细胞进行混合淋巴细胞培养时,反应细胞无3H-TdR掺入量增多(P>0.05);细胞周期蛋白依赖抑制因子(p16)阳性细胞数明显多于正常对照组及早期阶段。结论:这些变化可能与细胞周期紊乱。 Purpose To study the influnce of superantigen SEA on mice immnological cells. Methods SEA was injected into mice and the immunological cells were observed with immunological and morphological methods. Results SEA caused immunological cells proliferation early, which presented a high level of 3H TdR incorperation, and lymphocyte transformation, then apoptosis late in mice. The apoptotic cells showed karyopyknosis, karyorrhexis under microscopic and electromicroscopic, DNA in ladder appearance in electropheresis in 1 8% agrose. In the mixed lymphocyte reaction test, those cells had no stimulating effects on reacting cells, which showed no etra 3H TdR incorperation ( P>0 05 ). p16, a kind of inhibiter of cyclin dependent kinase was expressed higher in those cells than normal control and early ones. Conclusion The mechanism of those changes were not clear, which may be associated with the disorder of cell cycle and disturbance of cell signal transduction.
