摘要 目的:探讨Bim-1基因在胃腺癌细胞株、胃癌组织和癌旁组织以及胃癌患者外周血中的表达水平及其与临床病理特征的关系。方法:收集80例胃癌患者的癌组织和癌旁组织,培养胃腺癌细胞株(SGC-7901,BGC-823,AGS),采用RT-PCR和实时PCR的方法检测Bmi-1基因的表达情况。另外同时收集Bim-1基因阳性胃癌患者外周血标本以及健康体检者外周血标本各20例,用淋巴细胞分离液(Ficoll)分离获得单个核细胞,采用巢式PCR和实时PCR的方法检测其Bmi-1基因的表达情况。采用蛋白质印迹法验证胃癌组织和对应的癌旁组织以及胃癌患者及健康体检者血单个核细胞中Bim-1蛋白的表达情况。结果:胃腺癌细胞株(SGC-7901,BGC-823,AGS)中Bim-1均高表达;胃癌组织中Bim-1表达(85%)明显高于癌旁组织(30%)(P<0.05);胃癌患者外周血单个核细胞Bim-1基因的阳性率为60%,显著高于健康体检者(P<0.05)。胃癌组织和癌旁组织中Bim-1的蛋白水平存在差异,而在外周血中未见明显差异。结论:Bim-1基因在胃腺癌细胞株以及胃癌组织中高表达,胃癌患者外周血单个核细胞Bim-1的阳性率高于健康体检者,通过检测Bim-1基因可为胃癌的临床诊断及其靶向治疗提供新途径。 Objective:To detect the expression of Bim-1 gene in gastric carcinoma,adjacent non-cancerous tissues and the peripheral blood of the corresponding gastric carcinoma patient,to explore whether its expression is correlated with the clinical pathology of the gastric carcinoma.Methods:The expression of Bim-1 gene was detected by reverse transcriptase polymerase chain reaction(RT-PCR) and real-time PCR in gastric carcinoma,and adjacent non-cancerous tissues from 80 patients and the gastric carcinoma cell strain(SGC-7901,BGC-823,AGS).Furthermore,we detected at the same time the expression of Bim-1 gene in the peripheral blood of both 20 corresponding gastric carcinoma patients and health people.Using real-time PCR and nested RT-PCR detected the expression of Bim-1 gene in the peripheral blood.Western blot were employed to detect Bim-1 protein level.Results:The positive rates of Bim-1 expression was found in gastric tumor tissues was 85% (68/80),which was significantly higher than those was found in the matching,adjacent non-cancerous tissues 30% (24/80) (P0.05),A high level of Bim-1 expression was found in the gastric carcinoma cell strain(SGC-7901,BGC-823,AGS).In addition,The positive rates of Bim-1 expression in gastric carcinoma patient was higher than the healthy people.Bim-1 expression at the transitional levels were markedly different between gastric carcinoma and adjacent non-cancerous tissues by Western blot analysis.Conclusion:Bim-1 expression may play an important role in the development of the gastric cancer.It may be a novel diagnostic marker of human gastric cance.
