摘要
[目的]比较不同提取方法在金顶侧耳子实体中多糖的提取效果,建立金顶侧耳多糖含量的测定方法.[方法]分别采用热水浸提法和超声提取法提取多糖,利用苯酚-硫酸法和蒽酮-硫酸法测定金顶侧耳子实体中多糖的含量.[结果]超声提取法的得率较热水浸提法高;苯酚-硫酸法测定金顶侧耳多糖的平均回收率为98.69%,RSD为2.44%,蒽酮-硫酸法的平均回收率为99.69%,RSD为2.76%;同一个样品中苯酚-硫酸法和蒽酮-硫酸法所测得的多糖含量分别为7.34,9.49 g/kg.[结论]苯酚-硫酸法和蒽酮-硫酸法均可用于金顶侧耳中多糖含量的测定,但苯酚-硫酸法的稳定性和专属性优于蒽酮-硫酸法.
OBJECTIVE To compare the extracting effects of polysaccharids from the Pleurotus citrinipileatus Sing.bodies by different extraction methods,and to establish the detecting method for the polysaccharide content of the Pleurotus citrinipileatus Sing.bodies.METHODSThe polysaccharide was extracted by ultrasonic wave and hot-water extraction method,and the content of polysaccharide was detected by the phenol-sulfuric acid method and anthrone-sulfuric acid method.RESULTSThe yield of polysaccharide was better by ultrasonic wave method than by hot-water extraction method.The recovery of polysaccharides by phenol-sulfuric acid method was 98.69%,RSD was 2.44%,and the recovery of polysaccharides by anthrone-sulfuric acid method was 99.69%,RSD was 2.76%.The polysaccharide content at the same sample measured by the phenol-sulfuric acid method and anthrone-sulfuric acid method were 7.34 and 9.49g/kg,respectively.CONCLUSIONThe phenol-sulfuric acid method and anthrone-sulfuric acid method can be used to detect the polysaccharide content in the Pleurotus citrinipileatus Sing.bodies,and the stability and specificity of phenol-sulfuric acid method is better than that of anthrone-sulfuric acid method.
