缺氧诱导因子α亚型基因在急性心肌梗死大鼠缺血心肌表达的变化

摘要 目的通过观察急性心肌梗死(AMI)大鼠心肌缺氧诱导因子(hypoia-induciblefactor,HIF)3个α亚型mRNA表达的动态变化,探讨其在早期缺血心肌中的作用。方法将大鼠随机分为对照组和AMI组。用HE染色观察心肌形态学变化,RT-PCR检测术后1、2、4、6、12h缺血心肌HIF3个α亚型mRNA表达情况。结果HIF-1α、2α、3αmRNA在正常心肌中均有表达。术后1h,缺血心肌HIF-1αmRNA表达明显上调(P<0.05),2h达高峰(P<0.01),4h开始下降(P<0.05),6h降至正常水平,12h维持在正常水平;HIF-2α、3αmRNA在急性缺血心肌表达无明显变化。结论早期急性心肌缺血可诱导HIF-1α基因表达改变,与HIF-2α、3α基因表达无关。 Objective To investigate the expression regularity of hypoxia-inducible factor-α (HIF-α) mRNA in early ischemic myocardium.Methods The SD rats were randomly divided into control group and acute myocardial infarction (AMI) group.The ischemic myocardium was sampled for histological examination.RT-PCR was used to investigate the expression of HIF-α mRNA in ischemic myocardium at 1,2,4,6 and 12 h after operation.Results The expression of HIF-1α,-2α and-3α mRNA were all detectable in control group.HIF-1α mRNA began to elevate obviously in ischemic cardiac muscle at 1 h (P〈0.05),reached its peak at 2 h after operation (P〈0.01),decreased at 4 h (P〈0.05) and declined to normal level at 6 h.The expression of HIF-2α or-3α mRNA had no significant change compared with that of ischemic myocardium.Conclusions Early acute myocardial ischemia of rats can induce the change of expression of HIF-1α gene,which has no relation with the expressions of HIF-2α or-3α gene.
