低分子量肝素对E-选择素介导的HL-60细胞与血管内皮细胞黏附的影响

摘要 目的研究低分子量肝素(LMWH)对E-选择素介导的HL-60细胞与血管内皮细胞黏附的影响。方法通过原代培养方法分离人脐静脉内皮细胞(HUVEC)并建立体外模拟毛细血管流动的层流体系,利用该体系观察LMWH对体外动态条件下HL-60细胞与TNF-α诱导的HU-VEC的黏附作用的影响,荧光读板仪测定LMWH对E-选择素表达的影响。结果TNF-α处理HUVEC5h,能明显增加HL-60细胞与HUVEC在动态条件下黏附,并能明显促进HUVEC表面E-选择素表达。0.1、1mg/ml对HL-60细胞体外黏附有明显抑制作用;0.01、0.1、1mg/ml对脐静脉内皮E-选择素的表达有明显抑制作用。结论LMWH有明显抑制体外动态条件下HL-60细胞与人脐静脉内皮细胞黏附及降低血管内皮细胞E-选择素表达的作用。这一结果为LMWH作为抗炎药物,尤其是为以选择素作为药物作用靶点应用于临床提供了重要的实验基础。
