乳腺良恶性肿瘤及乳腺癌细胞系中SSEA-3和SSEA-4的表达

Expressions of stage specific empryonic antigen-3 and-4 in benign and malignant breast tumors as well as in MDA-MB-468 breast cancer cell line

摘要 目的:探讨阶段特异性胚胎抗原SSEA-3及阶段特异性胚胎抗原SSEA-4在乳腺良恶性肿瘤及乳腺癌细胞系MDA-MB-468中的表达情况,及其与乳腺癌发生的关系。方法:通过免疫组织化学和免疫细胞化学方法检测50例乳腺癌(浸润性导管癌)病变、50例乳腺良性病变(纤维腺瘤,乳腺腺病)及乳腺癌细胞系MDA-MB-468中SSEA-3和SSEA-4的表达。结果:SSEA-3在乳腺癌中的表达阳性率为34%(17/50),且SSEA-3在乳腺癌中的表达与ER表达呈正相关(P<0.05),而在乳腺良性病变中的表达阳性率为16%(8/50);SSEA-4在乳腺癌中的表达阳性率为58%(29/50),在乳腺良性病变组织中表达阳性率为36%(18/50);SSEA-3和SSEA-4在乳腺癌中的表达均高于其在良性病变中的表达(P<0.05);且两者在MDA-MB-468细胞系中均有阳性表达。结论:SSEA-3和SSEA-4在乳腺癌良恶性病变中的表达有区别,而且在乳腺癌细胞系MDA-MB-468中表达阳性率较高,提示SSEA-3和SSEA-4的表达可能与乳腺癌的发生有关。 OBJECTIVE:To investigate the expressions of SSEA-3 and SSEA-4 in benign and malignant tumors of the breast and in breast cancer cell line MDA-MB-468,as well as their relationship with the breast carcinogenesis.METHODS:Expressions of SSEA-3 and SSEA-4 were immunohistochemically/immunocytochemicaaly investigated in 50 benign(fibroadenoma,adenosis of breast) and 50 malignant(infitrating ductal carcinoma) breast tumor samples,in addition to the breast cancer cell line MDA-MB-468.RESULT:The positive rate of SSEA-3 in breast carcer was 34%(17/50),and it was also discovered that SSEA-3 expression was significantly associated with positive ER expression in the malignant samples(P〈0.05).Besides,the positive rate of SSEA-3 in benign breast tumor was 16%(8/50);However,the positive rate of SSEA-4 in breast carcer was 58%(29/50),and its positive rate in benign breast tumor was 36%(18/50);Both SSEA-3 and SSEA-4 showed a higher level expression in the malignant tusamples compared to the expression in the benign ones(P〈0.05).Besides,both SSEA-3 and SSEA-4 expressions were detected in the breast cancer cell line MDA-MB-468.CONCLUSIONS:The expression of SSEA3 and EESA4 were different between in the benign and malignant breast tumor.Besides,they both show high expression in the breast cancer cell line MDA-MB-468,indicating that these proteins may be linked to breast cancer development.
