摘要
以3周龄乌鸡腿肌为试验材料,提取RNA进行RT-PCR反应、TA克隆和测序,并进行序列测定。结果表明:乌鸡腿肌中存在3种不同的MSTN基因转录子,分别将其命名为cbMSTN-1、cbMSTN-2和cbMSTN-3;cbMSTN-1由1128bp组成,cbMSTN-2由985bp组成,cbMSTN-3由754bp组成;cbMSTN-2与cbMSTN-1相比缺失了374~517bp处的143bp碱基,cbMSTN-3与cbMSTN-1相比缺失了374~748bp处的374bp碱基;cbMSTN-2、cbMSTN-3与cbMSTN-1的同源性均为99%。
RT-PCR was applied in this experiment to study the expression of myostatin. It was found that there were three MSTN gene transcriptons of varied sizes in muscle of blackchicken by cloning and sequencing. Then they were named cbMSTN-1,cbMSTN-2 and cbMSTN-3. cbMSTN-1 was composed of 1 128 bases; cbMSTN-2 was composed of 985 bases; cbMSTN-3 was composed of 754 bases. Compared with cbMSTN-2 and cbMSTN-1, 143 bases were deleted from 374 to 517; compared with cbMSTN-3 and cbMSTN-1, 374 bases were deleted from 374 to 748; compared with cbMSTN-3and cbMSTN-2, 231 bases were deleted from 374 to 748. cbMSTN-1 held 99% homology with cbMSTN-2,four bases were changed. It held 99% homology with cbMSTN-3, six bases were changed; cbMSTN-2 held 99% homology with cbMSTN-3,,four bases were changed. The results provided experimental basis to study the mechanism of MSTN gene.
