摘要
试验研究了经BHK-21培养的口蹄疫病毒(FMDV)OS/99株VP1蛋白的分子免疫学特征。通过RT-PCR得到FMDV的VP1基因序列,应用分子生物学软件对其蛋白序列的空间构型和抗原性进行分析。通过对其β-折叠数量、亲水性、抗原指数、可塑性、表面可能的抗原性参数的综合分析,得出OS/99株VP1蛋白中可能含有多个抗原位点,具有较强的抗原覆盖性。抗原位点可能位于22-29、43-46、82-85、92-93、100-111、134-139、154-156、180-184和196-203位氨基酸残基区域内。毒株经过BHK-21细胞培养13代后,其蛋白空间构型未发生明显变化。
The aim of our experiment was to investigate molecular immunological characters of VP1 in foot-and-mouth disease virus from OS/99 viral strain cultured in BHK-21 cells . In this study,the sequence of VP1 ,which was isolated through RT-PCR , was analyzed in DNAstar software to abtain its spatial arrange and antigenicity . The result of this research indicated that the VP1 from OS/99 might contain many antigenic sites in terms of some parameters on the antigenicity , which located in 22-29,43-46,82-85,92-93,100-111,134-139,154-156,180-184,196-203 amino acid regions . For that , the virus strain of os/99 showed the powerful ability in aspect of more broader coverage in antigenicity Together ,it was suggested that the spatial conformation of VP1 in OS/99 virus strain ,which were subcultured for 13 generations , have not been obviously changed .
