摘要
目的:优选白术内酯Ⅰ脂质体最佳制备工艺及处方。方法:采用薄膜分散法制备白术内酯Ⅰ脂质体,以包封率为评价指标,采用正交设计优化处方和制备工艺,采用HPLC法测定其包封率。结果:最佳处方工艺为:卵磷脂与胆固醇的质量比为6:1,卵磷脂与白术内酯Ⅰ质量比为20:1,PBS缓冲溶液pH为7.5。结论:经优化得到的白术内酯Ⅰ脂质体处方合理,工艺可行,包封率较高。
Objective： To optimize the prescription and preparation of atractylenolide I liposomes. Method： Atractylenolide I lipo- somes were prepared by film dispersion method. The encapsulation efficiency was taken as inspection target and the preparation of lip- somes were optimized by orthogonal design. HPLC was used to determine the encapsulation efficiency. Result： The best prescription was as follow：the ratio of egg phospholipids to cholesterol and egg phospholipids to atractylenolide I was 6：1 and 20： 1, respectively, and the pH of phosphate buffer solution was 7.5. Conclusion： The optimized formulation of atractylenolide I liposomes is reasonable in pre- scription, practicable in technology, high in encapsulation efficiency.
