中药复方抗纤抑癌方和复方鳖甲软肝片不同采血时间点药物血清对HepG2肝癌细胞增殖活性的影响

Influence of Kangxian yiai Decoction and Fufang Biejia Ruangan Tablets on HepG2 Cells on Different Blood Sampling Spots

目的:研究人来源不同采血时间点中药药物血清对HepG2人肝癌细胞增殖活性的影响。方法:HepG2人肝癌细胞体外培养,健康志愿者3名,服用中药复方抗纤抑癌方(KX)和中成药复方鳖甲软肝片(BJ)后不同时间点收集血清,采用MTT法检测各组药物血清对细胞增殖的影响。结果:(1)1h、2h与MEM对照组比较,各实验组药物血清对HepG2人肝癌细胞具有明显的增殖抑制作用(P<0.01);KX1h20%浓度抑制效果优于KX2h同浓度(P<0.01);KX1h5%浓度与BJ1h同浓度有差异(P<0.05);BJ2h20%、10%浓度均较KX2h抑制作用强(P<0.05);(2)增加0.5h抽血时间点,KX0.5h优于KX2h(P<0.05);(3)与空白血清组比较,KX0.5h40%浓度时抑制效果明显(P<0.05)。结论:2种药物血清对HepG2人肝癌细胞均有增殖抑制作用;在同一采血时间点和不同时间点上,2种药物血清的抑制效果比较均存在差异。

Objective： To explore the influence of different kinds of human-derived chinese medicine sera on proliferation of HepG2 cells on different blood sampling spots.Methods： HepG2 cells were cultured in vitro.The sera of 3 volunteers were collected before taking medicine as the control group,twice after taken medicine（Kangxian yiai decoction and Fufang Biejia Ruangan Tablets） 1 hour later and 2 hours again as the test group,MTT colorimetric assay was used to observe the influence on proliferation of HepG2 cells after incubate with different kinds of drug＇s containing serum.Results： 1.It showed that The inhibited proliferation in test groups were better than that of control group（P 0.01）.The inhibited proliferation of the concentration 20% of KX were better after 1hour than that of 2hours,and the difference had statistical significance.Compared with 5% KX group on the 1-hour blood sampling spot,the inhibited proliferation of the concentration 5% of BJ were better,and the difference had statistical significance（P 0.05）.While on the 2-hour blood sampling spot,the inhibited proliferation of different concentration of BJ were better than that of KX,the differences in BJ groups of 20% and 10% concentration had statistical significance （ P 0.05）.2.In KX groups,the inhibited proliferation of different concentrations on HepG2 cells were better on the half-an-hour blood sampling spot than 2hour,and the difference had statistical significance （ P 0.05 ）.3.The inhibited proliferation of 40% concentration on the half-an-hour blood sampling spot in KX group were better than that in control group,and the difference had statistical significance （ P 0.05）.Conclusion： Both the two kinds of chinese herbal compound sera can inhibit the proliferation of HepG2 cells.The inhibited proliferation of different concentrations on HepG2 cells were better on the half-an-hour blood sampling spot than 2 hour.It showed the trend that the inhibitory effects of the sera in BJ group on 2-hour blood sampling spot were better than 1 hour.The inhibitory effects of the two kinds of sera on different blood sampling spots or on the same spot were different.
