孕哺期母鼠镧暴露对子代神经系统的影响 被引量:1

Effect of Lanthanum Chloride Exposure in Rats during Pregnancy and Lactation on Central Nervous System of Their Offspring

摘要 目的探讨孕哺期母鼠镧暴露对子代神经系统的影响。方法Wistar母鼠从妊娠起至仔鼠断乳止通过饮水连续染镧21 d,跳台实验检测仔鼠学习记忆能力,荧光分光光度计法测定细胞内Ca2+浓度,流式细胞仪法测定海马细胞凋亡率。结果随着三氯化镧(LaCl3)剂量的增加,镧暴露子代大鼠潜伏期显著下降,而错误次数显著增加;细胞内钙离子浓度([Ca2+]i)显著升高;镧暴露组海马细胞凋亡率显著增加,而且差异均有统计学意义(P<0.05)。结论孕哺期母鼠镧暴露可以损害子代大鼠学习记忆功能,可能与镧暴露使海马细胞内游离Ca2+浓度升高和细胞凋亡增加有关。 Objective To explore the effect of lanthanum chloride exposure in rats during pregnancy and lactation on central nervous system of their offspring.MethodsThe female rats were administrated with lanthanum from pregnancy to the end of lactation.Platform experiment was used to detect the activities of neural behavior including learning and memory.Fura-2/AM calcium ions fluorescence indicator was used to measure Ca^2+ concentration in hippocampal neurons.FAC method was adopted to measure the apoptosis level in hippocampus cell of their offspring.ResultsAs the exposure dosages increased,the incubation of the offspring shortened significantly,accompanied with obvious increase in mistake times,and distinctive elevation of Ca^2+ concentrations in cells and apoptosis levels.The differences were significant(P〈0.05).ConclusionsLanthanum exposure in rats during pregnancy and lactation may harm the functions of learning and memory of their offspring.The causation may be associated with the elevation of the concentration of Ca^2+ and the increase of apoptosis.
