摘要 应用Bcl-2反义硫代磷酸寡核苷酸(ASPO)与HL60细胞共培养,旨在观察不同浓度ASPO对HL60细胞生长和克隆形成的影响。结果发现,与Bcl-2正义寡核苷酸(SPO)及空白组比较,不同浓度的ASPO(A5μmol/L,A10μmol/L,A20μmol/L)培养24小时即可降低HL60细胞生长数和CFU-HL60克隆形成率。并随浓度的增大而显著(P<0.01)。72小时以后,ASPO组则与SPO组和空白组无显著差别(P>0.05)。因此认为Bcl-2的ASPO具有抑制HL60细胞生长,并呈浓度依赖性和序列特异性。 To observe the effect of the BCL-2 antisense phosphorothioate oligodeoxynucleotides (ASPO) inhibition on HL60 cells growing, we investigated the cell viability determined by trypan blue exclusion and the colony forming activity (CFU-HL60) after incubation of HL60 celis with ASPO at different concentration (5μmol/L, 10μmol/L, 20μmol/L) or with sense-PO(SPO) at 20μmol/L or with none of them. The result demonstrated that in the presence of ASPO, the inhibition of HL60 cells viability could occur in 24 hours and became more prominent as the concentration increas-ing (P<0. 01). That was not found in the presence of SPO. There was no significant differences among ASPO, SPO and the control after 72 hours. These findings indicated that BCL-2 ASPO could inhibit the HL60 cells proliferation and this effect was sequences-specific and concentration-dependent.
