环氧化酶-2和血管内皮生长因子在人膀胱移行细胞癌中的表达及对血管形成的影响

Expression of COX-2 and VEGF in Human Transitional Cell Carcinoma of Bladder and Theirs Influence on Angiogenesis

摘要 目的检测环氧化酶-2(COX-2)、血管内皮生长因子(VEGF)和CD105在人膀胱移行细胞癌(TCCB)组织和正常膀胱组织中的表达,探讨COX-2和VEGF的表达与血管形成的关系。方法采用RT-PCR法检测人TCCB和正常膀胱组织中COX-2 mRNA的表达。采用免疫组织化学法检测人TCCB和正常膀胱组织中VEGF和CD105的表达并对微血管进行计数。结果正常膀胱组织不表达COX-2 mRNA,VEGF和CD105的表达很低;人TCCB组织中COX-2 mRNA、VEGF和CD105的表达均显著高于正常膀胱组织(P<0.01或<0.05)。低分化和肌层浸润型癌组织中COX-2 mRNA、VEGF和CD105表达显著高于高分化和非肌层浸润型癌组织(P<0.01或<0.05)。结论COX-2和VEGF与人TCCB的分级和分期有关。COX-2和VEGF与癌组织中血管形成密切相关。 Objective To investigate the expressions of cyclooxyenase-2 (COX-2) , vascular endothelial grouth factor(VEGF) and CD105 in tissues of normal human bladder and the human bladder with transitional cell carcinoma respectively, and the effect of the expressions of COX-2 and VEGF on angiogenesis. Methods The expressions of COX-2 mRNA in tissues of normal human bladder and the human bladder with transitional cell carcinoma were tested by RT-PCR. Then the expressions of VEGF, CD105 and the quantity of angiogenesis in tissues of normal human bladder and the human bladder with transitional cell carcinoma were detected by immunohistochemistry. Results All normal bladder tissues show negative expression of COX-2 mRNA and low expressions of VEGF and CD105, while the expression of COX-2 mRNA, VEGF and CD105 in the tissues of the human bladder with transitional cell carcinoma was significantly higher than those in normal bladder tissues(P 〈 0.01 or 〈 0.05 ). The expressions of COX-2 mRNA, VEGF and CD105 in poorly-differentiated and invasive bladder carcinoma tissues was significantly higher than those in well-differentiated and superficial carcinoma tissues (P 〈 0.01 or 〈 0.05 ). Conclusion The expression of COX-2 and VEGF are closely related to the tumor stage, grade and angiogenesis.
