摘要
目的探讨脱氢表雄酮对大鼠血管平滑肌细胞活性及细胞周期的影响。方法培养大鼠胸大动脉血管平滑肌细胞系,血小板源性生长因子(PDGF)刺激细胞增殖,采用CCK-8方法观察不同浓度脱氢表雄酮对血管平滑肌细胞活性的影响,用流式细胞分析法检测细胞周期分布。结果1.0、2.5、5.0μg/LPDGF增强大鼠血管平滑肌细胞活性,0.1、1.0、10、100nM脱氢表雄酮抑制细胞活性,两者的作用随浓度增加而增强;脱氢表雄酮可抑制PDGF的作用。脱氢表雄酮的作用增加G1期的细胞数量。结论脱氢表雄酮通过抑制细胞周期的进展,而减少血管平滑肌细胞的增殖。
Objective To investigate the effects of dehydroepiandrosterone(DHEA)on the viability and cell cycle of vascular smooth muscle cells (VSMCs).Methods VSMCs were cultured and stimulated with platelet-derived growth factor (PDGF),effects of DHEA at different concentrations on viability of VSMC were studied by means of CCK-8 method,and cell cycle distribution was analyzed using flow cytometry.Results PDGF(1.0、2.5、5.0 μg/L) increase while DHEA (0.1、1.0、10、100 nM) suppress cell viability,the effect of PDGF could be disabled by DHEA. The inhibition of PDGF-stimulated VSMC proliferation by DHEA was accompanied by an increase in G1-phase population.Conclusion DHEA inhibited PDGF-induced VSMC growth via inhibition of cell cycle progression.
