摘要
目的:探讨香茶菜属植物冬凌草的主要有效成分冬凌草甲素提取纯化方法。方法:以HPLC法测定提取物中冬凌草甲素的含量为检测指标,考察影响冬凌草甲素提取分离的各种相关因素。结果:研究表明,按10 L/kg干草的比例使用80°C乙醇浸泡提取30 min得到的冬凌草浸膏是适合实验室大规模提取冬凌草甲素的方法,采用石油醚丙酮溶剂体系进行首次硅胶柱层析分离可以去除浸膏中大部分杂质,采用乙酸乙酯乙醇体系进行二次硅胶柱层析分离并加以乙醇结晶处理可以得到99%以上纯度的冬凌草甲素。结论:确定了适合实验室内较大规模提取冬凌草甲素的方法。
Objective: To explore a suitable extraction and purification method for deriving ordonin from Rabdosia rubbescens.Methods: The contents of oridonin was determined by HPLC,different methods and related factors in extraction and purification were presented.Results: The research shows that at the proportion of 10 L/kg hay,incubate 80 ℃ for 30 min immersion extract,the obtained oridonin extract was suitable for large-scale laboratory extraction.Petroleum ether and acetone solvent system for the first time silica column chromatography can remove most of extract impurities.Ethyl acetate and ethanol solvent system for second silica column chromatography with crystallization in ethanol can obtain more than 99% high purity oridonin.Conclusion: The method for deriving high purity oridonin on a large scale in laboratory was determined.
