骨髓间充质干细胞体外诱导分化为心肌样细胞 被引量:11

摘要 目的:探讨肝细胞生长因子(hepatocyte growth factor,HGF)作为诱导剂,体外诱导大鼠骨髓间充质干细胞(mesenchymal stem cells,MSCs)分化为心肌细胞的作用。方法:分离培养大鼠MSCs,在第4代MSCs中添加HGF(20ng/mL)对其进行诱导分化,观察细胞形态学的变化,采用荧光免疫组织化学及RT-PCR方法对分化细胞进行鉴定。结果:大鼠MSCs贴壁呈集落生长,有成纤维细胞样外观。诱导后细胞呈梭形,排列方向渐趋一致,有肌管样结构形成,未观察到转化细胞的自主搏动。Troponin T及Connexin43的表达呈阳性,并可见清晰肌小节。RT-PCR检测MLC-2A、MLC-2V、α-MHC、β-Actin mRNA表达阳性。结论:大鼠MSCs体外在HGF诱导下可定向分化为心肌样细胞。
