摘要
以单克隆抗体AH6[特异结合LeY寡糖,Fucα12Galβ14(Fucα13)GlcNAc]以及其它3种与LeY寡糖结构近似的寡糖特异结合的单克隆抗体αLea、αLeb和FEA5为工具,采用小鼠体外着床模型,研究细胞表面LeY寡糖抗原与着床的关系及其在着床过程中的具体作用环节。结果显示:不论子宫上皮细胞表面还是胚胎滋养层细胞表面的LeY寡糖抗原被封闭后,都能导致胚胎在单层子宫上皮细胞表面粘附和扩展生长的延迟,但对胚胎最终的扩展程度并无影响,而其它3种单克隆抗体对胚胎的粘附则均无影响。说明这种作用具有明确的寡糖结构特异性。另外,运用免疫荧光方法观察到,胚胎在单层子宫上皮细胞表面的粘附与LeY寡糖在细胞表面的消长密切相关。因而认为LeY寡糖抗原在胚胎着床中可能是作为一种中介分子参与着床始发阶段的胚胎与母体间的识别和粘着。实验中经单克隆抗体处理后胚胎和单层子宫上皮细胞与对照组比较,其形态与发育未见不同。
The Chinese Academy of Sciences, Beijing 100080 , China) ( ②Department of Biochemistry, Dalian Medical University, Dalian 116027, China) To study the action of glycoantigen specified expression on the surface of embryos and uterine epithelial cells on implantation, monoclonal antibody AH 6 [against Le Y oligosaccharide, Fuc α1 2Galβ1 4(Fuc α1 3)GlcNAc ] and other three monoclonal antibodies αLe a, αLe b, FE A 5 against oligosaccharides with structures similar to Le Y oligosaccharide were used. By mouse implantation model in vitro , the results showed that blastocyst attachment and outgrowth at the surface of uterine monolayer epithelial cells were all postponed whether cell surface Le Y glycoantigen was blocked on the epithelial cells or on the trop hoblasts, but there was no effect on embryo′s outgrowth at the end. No any effect was observed with the other three monoclonal antibodies; By immunofluorescent method,it was observed that blastocyst attachment to the surface of uterine epithelial cells was closely related to the existence of Le Y glycoantigen. It was, therefore, concluded that oligosaccharide Le Y plays an essential role at the initial stage of implantation. It may act as a mediator molecule for the recognition and adhesion between the surface of blastocyst and epithelial cell, and its function is carbohydrate specific. The oligosaccharide consisting of two fucose groups linked with H type 2 chain through α1 2 and α1 3 linkages may be the determinant of the molecule. Monoclonal antibodies used in this study have no side effect on the cell.
