摘要
为探讨老年性记忆减退的机制,将老年大鼠分为老年记忆减退组(简称老年减退组)和老年记忆正常组(简称老年正常组)。用免疫细胞化学、组织化学双标技术定量分析内侧隔核-斜角带(SM-DB)中神经生长因子受体(NCFR)和NADPH-d阳性神经元的数量、其结果是:老年减退组大鼠和老年正常组大鼠相比较,SM、斜角带垂直支(vDB)、水平支(hDB)中的NGFR/NADPH-d双重反应阳性、NGFR单独反应神经元数也呈明显下降;NADPH-d单独反应阳性神经元的数量也较老年正常组减少(P<0.05)。相关分析表明,受试大鼠逃避潜伏期与双重反应阳性神经元、NGFR和NADPH-d单独反应阳性神经元的数量呈明显负相关。提示SM-DB的三类神经元都参与了老年记忆减退的机制,但以NGFR阳性神经元的损害起主要作用。
To explore the mechanism of senile memory deficits, 60 male healthy SD rats, including 20 young (3 months) and 40 old (26 months) were examined with Morris water maze. The old were divided into two groups, senile memory deficits rats (SMD) and senile normal memory rats (SNM). The subpopulations of nitric oxide synthasc (NOS) and nerve growth factor receptor (NGFR) neuron of SM-DB complex were analyzed qualitatively and quantitatively. There were 3 kinds of neurons (NGFR/NOS double labeled, NGFR single labeled and NOS single labeled) in SM-DB complex of SMD and showed significant difference ascompared with that of SNM. That of SNM and young showed no significant difference. The reduction of NGFR single labeled neurons in the SM-DB of SMD was most obvious. The decrease range of the NOS single labeled neuron of SMD was smaller than that of others. There were significant correlation between the mean escape latency of the examined rats and the number of double labeled, the NGFR single labeled, and NOS single labeled neurons. Although the double labeled, the NGFR labeled and NOS single labeled ncurons decreased in SM-DB complex of SMD, but the meanings were different. The NGFR and NOS subpopulation played different roles in the mechanism of memory. The loss of the NGFR single labeled neuron was correlated closely with senile memory deficits.
