摘要 目的采用曲线拟合方案测定单克隆抗体亲和常数计算中的OD100。方法以测定抗苏丹红-1单克隆抗体亲和常数为例,非竞争ELISA法测定OD值,经曲线回归拟合结合反应的曲线模型及其方程,依据方程计算OD100值。结果根据幂曲线模型计算的抗苏丹红-1单抗亲和常数为K=8.19×108l/mol。结论应用统计学分析方法拟合的曲线模型,科学可靠地测定了OD100,为单克隆抗体亲和常数的计算提供了科学、合理的测量基础。 Objective To calculate ODloo in measurement of affinity constant of monoclonal antibody. Methods The affinity constant of monoelonal antibodies against sudan-1 was determined by noncompetitive enzyme immunoassay with curve-fitting method. Results Power model was fitted based on scatter diagram of the binding reaction of antigen and antibody. Conclusion OD100 was determined credibly by application of statistical methods which provided a rational measurement for calculating affinity constant of monoclonal antibody.
