摘要 目的探讨正常肝组织与乙型肝炎中TAP1、TAP2、HSP70、HSP90、CNX、β2-MG蛋白表达及相关关系。方法以内源性生物素阻断S-P法来检测32例乙肝患者和16例健康对照者石蜡包埋肝组织的TAP1、TAP2、HSP70、HSP90、CNX、β2-MG蛋白表达情况。结果β2-MG、TAP1、HSP70、HSP90、CNX蛋白在乙肝组表达高于正常组(P均<0.05);通过相关性分析发现,TAP2蛋白与HSP70蛋白在正常组中呈相关性(χ2=16.000,P=0.008)。结论TAP1、TAP2、HSP70、HSP90、CNX、β2-MG蛋白均在乙肝组表达高于正常组(P<0.05),提示这些蛋白均参与MHC-Ⅰ类分子的乙肝病毒抗原处理及提呈过程,并且可以做为诊断乙肝的临床指标。 Objective To evaluate the expression levels of the MHC class Ⅰantigen presentation-associated proteins: transport associated antigen-processing proteins TAP1 and TAP2,heat shock protein(HSP)70 and HSP90,calnexin(CNX),and β2 microglobulin(β2-MG),in normal and hepatitis B liver tissues,and its association with hepatitis B.Methods The expression of proteins TAP1,TAP2,HSP70,HSP90,CNX,and β2-MG in paraffin-embedded liver tissues from 32 patients with hepatitis B(as hepatitis B group) and 16 healthy individuals(as control group) were detected by streptoavidin-biotin-peroxidase(S-P) based immunohistochemistry method with blocking endogenous biotin.Results The expression level of β2-MG,TAP1,HSP70,HSP90 and CNX in liver tissues from hepatitis patients was higher than that in normal group(P〈0.05).The correlation analysis suggested a correlative relationship between expression of TAP2 and HSP70 in the normal liver tissues(χ^2=16.000,P=0.008).Conclusion The expression of TAP1,TAP2,HSP70,HSP90,CNX,and β2-MG proteins was higher in the hepatitis B group than that in the normal group(P〈0.05).This results suggest that these proteins are involved in the process and presentation of MHC class Ⅰ antigens of hepatitis B virus,and could be used as markers for clinical diagnosis of hepatitis B.
