辛芷鼻敏胶囊对变应性鼻炎大鼠脾脏组织中STAT6 mRNA表达变化 被引量:1

摘要 目的探讨辛芷鼻敏胶囊对变应性鼻炎(Allergic rhinitis,AR)大鼠脾脏组织中的STAT6 mRNA的表达变化。方法将40只大鼠随机分为空白组、模型组、对照组、实验组,每组10只,用卵清蛋白建立AR大鼠模型,通过对造模的大鼠灌服辛芷鼻敏胶囊(2.3g.kg-1.d-1)及与同类药物鼻炎康(1.8g.kg-1.d-1)对照,用原位杂交方法检测脾脏组织中STAT6 mRNA的阳性表达。结果实验组大鼠脾脏组织中STAT6 mRNA阳性表达量(0.162±0.005)明显低于模型组(0.191±0.006)及对照组(0.177±0.006),差异有统计学意义(P<0.05)。实验组大鼠变应性鼻炎症状明显减轻,仅有轻微抓鼻动作、喷嚏;实验组大鼠鼻黏膜组织病理学明显改变,腺体增生减轻,嗜酸性粒细胞浸润减少。结论辛芷鼻敏胶囊可能降低脾脏组织中STAT6 mRNA阳性表达,改善鼻粘膜嗜酸性粒细胞浸润而控制变应性鼻炎症状。 Objective To explore the effect of Xinzhi Bimin capsule on the express of STAT6 mRNA in spleen of rat models with allergic rhinitis.Methods Forty rats were divided into four groups at random as normal group,model group,control group and experimental group,each group were ten rats.Ovalbumin sensitized rats used as animal of AR were taken orally with Xinzhi Bimin capsule(2.3 g·kg^-1·d^-1)and compared with BiYan kang(1.8 g·kg^-1·d^-1),the expression of positive cell of STAT6 mRNA in spleen was detected in semi quantity by in situ hybridization(ISH).Results The expression of the proportion of positive cell to STAT6 mRNA in spleen were markedly lower in the experimental group(0.162±0.005)than the model group(0.191±0.006)and the control group(0.177±0.006)(P0.05),except the normal group.The symptom of rats with AR from experimental group markedly lower than that from other groups,which have only slightly scratch nose and sneeze;Gland hyperplasia and infiltration of eosinophil in nasal mucosa in the experimental group were reduced compared to that in model group and control group.Conclusion The result indicates that the capsule of Xinzhi Bimin can reduce the expression of STAT6 mRNA in spleen of rat models with allergic rhinitis and reduce the infiltration of eosinophil whereby to treat AR in rats.
