复方胡芦巴对小鼠乙醇肝损伤保护作用的实验研究

Experimental study on effect of compound Trigonella foenum-graecum. L protection against alcohol liver injury in mice

目的探讨复方胡芦巴对乙醇所致小鼠化学性肝损伤的保护作用。方法将84只雄性小鼠随机分成肝损伤模型组、正常对照组、阳性组(联苯双酯150mg/kg)、复方胡芦巴低(0.9g/kg)、中(1.8g/kg)、高(2.7g/kg)剂量组,每组14只。经预防性给药10d后,采用乙醇复制小鼠急性肝损伤模型,16h后眼眶取血,测定小鼠血清中谷丙转氨酶(GPT)、谷草转氨酶(GOT)含量及肝组织中丙二醛(MDA)、还原型谷胱甘肽(GSH)、甘油三酯(TG)的含量。观察肝脏病理组织学变化。结果复方胡芦巴高剂量组能明显降低乙醇所致肝损伤动物模型的血清GPT、GOT含量(P<0.05)。复方胡芦巴中、高剂量组肝组织中MDA含量低于模型组,而GSH含量高于模型组,经比较差异有统计学意义(P<0.05),胡芦巴可改善乙醇引起的肝脏组织的病理变化,对肝组织中TG变化无影响(P>0.05)。结论复方胡芦巴对乙醇所致小鼠肝损伤具有明显的保护作用。

Objective To explore the protective effects of compound Trigonella foenum-graecum.L against alcohol induced acute liver injury in mice.Methods Eighty-four mice were randomly divided into liver injury model group,negative control group,positive control group（Bifendate 150 mg/kg）,low（0.9 g/kg）,middle（1.8 g/kg）and high（2.7g/kg）dosage group of compound Trigonella foenum-graecum.L.Each group had 14 mice.Pretreated with compound Trigonella foenum-graecum.L for 10 days,mice model of the acute liver injury were established by the treatment of alcohol,16 hours later,the protective effects of compound Trigonella foenum-graecum.L in acute liver injury induced by alcohol were observed through determining the activiteds of GPT and GOT in serum;The contents of MDA,GSH and TG in its＇ liver tissues were determined;Histopathological changes of liver were also observed.Results Compound Trigonella foenum-graecum.L significantly decreased the contents of serum GPT and GOT in alcohol induced liver injury model mice（compared with model group,P〈0.05）;The contents of MDA in tested sample were higher than those of model group,but just reversed for GSH（P〈0.05）contents;The pathologica changes induced by alcohol in liver tissues were improved.There was no significant difference in the effects of TG（P〉0.05）.Conclusion Compound Trigonella foenum-graecum.L plays the protective effects obviously on acute liver injury induced by alcohol in mice.
