三氧化二砷对慢性粒细胞白血病骨髓间质干细胞活性的影响 被引量:5

摘要 目的探讨三氧化二砷(ATO)体外对骨髓间质干细胞(BMMSCs)的作用及其对慢性粒细胞白血病(CML)的治疗作用。方法体外培养法分离CML患者BMMSCs,培养体系中加入ATO,ATO终浓度分别为0、1.0、2.5、5.0μmol/L,MTT法测定BMMSCs生长抑制率;裂解BMMSCs并提取总RNA,Real-time PCR法测定其干细胞因子(SCF)基因表达水平并进行方差分析比较。结果2.5、5.0μmol/L ATO浓度组抑制率大于1.0μmol/L ATO浓度组(P均<0.05),1.0、2.5μmol/L ATO浓度组SCF基因表达水平较空白对照组显著升高(P均<0.01),1.0、2.5μmol/L ATO浓度组SCF基因表达水平比较差异亦有统计学意义(P<0.01)。结论ATO显著抑制BMMSCs生长活性,低浓度ATO促使BMMSCs高表达SCF,从而拮抗ATO的诱导凋亡作用;高浓度ATO使BMMSCs表达SCF降低,有利于细胞凋亡。
