仙鹤草乙醇提取物在急性肺损伤中的作用研究 被引量:4

摘要 为探讨仙鹤草乙醇提取物在脂多糖诱导急性肺损伤中的作用,使用NIH雄性小鼠40只,随机分为机分为空白组、模型组与实验组(相当于仙鹤草生药8g/kg组,16 g/kg组和24 g/kg组),每组8只。试验组以仙鹤草乙醇提取物连续灌胃(qd×5 d),空白组和模型组以等量N.S.灌胃。末次灌胃后以脂多糖(LPS)2.5 mg/kg气管滴注建立急性肺损伤模型。建模24 h后以酶联免疫法检测血浆前列腺素E2(PGE2)、白介素-10(IL-10)和环氧化酶-2(COX-2)含量。结果与模型组相比,仙鹤草乙醇提取物8 g/kg组,16 g/kg组和24 g/kg组血浆PGE2和COX-2水平显著降低(P<0.05)。与模型组相比,仙鹤草乙醇提取物8 g/kg、16 g/kg、24 g/kg组血浆IL-10水平显著升高(P<0.05)。结论仙鹤草乙醇提取物可能通过抑制COX-2表达,降低PGE2生成,并促进IL-10的生成对急性肺损伤产生保护作用。 Aim To investigate the effects of Agri.monia pilosa Ledeb ethanol extract on acute lung injury(ALl) induced by lipopolysaccharides(LPS). Methods Male mice were divided into sham operation group, nmdel group, Agri.monia pilosa Ledeb ethanol extract 8 g/kg group,16 g/kg group and 24 g/kg group randomly. Research groups were given Agri.monia pilosa Ledeb ethanol extract (ig, qd) for 5 days. Sham operation group and model group were given normal sodium(NS). Once the last dosage was given, the ALI model was built by intratracheal instillation of LPS 2.5 mg/kg. Mice were sacrificed 24h later and the content of prostaglandin E2 (PGE2), interleukin-10(IL-10) and cycloxygenase-2(COX-2) were detected by ELISA. Results Compared with model group, the content of PGE2 and COX-2 of all the Agri.monia pilosa Ledeb ethanol extract groups were decreased significantly(P〈0.05). The content of IL-10 of each Agri.monia pilosa Ledeb ethanol extract group was increased significantly compared with model group. Conclusion Agri.monia pilosa Ledeb ethanol extract may be benefit to the ALI by inhibiting the expression of COX-2, reducing the production of PGE2, and promoting the generation of IL-10.
