摘要
目的观察阴道毛滴虫在粘附状态下的形态结构特点。方法将固定液注入感染阴道毛滴虫的大鼠阴道,达到局部预固定效果后,再取材浸泡固定,分别进行透射电镜、扫描电镜、光镜免疫组化及特染观察。结果经预固定后制作的标本片虫体数量较多。电镜下见阴道毛滴虫呈阿米巴样粘附于上皮细胞表面,虫体指状伪足插入上皮细胞间。结论采用原位加浸泡固定方法制备的样品,虫体数量多,且更接近其粘附状态下的形态结构特点。
Objective To observe the characteristic morphological structures of Trichomonas vaginalis during adhesion.Methods A fixative solution was injected into rat vaginas infected with T.vaginalis to achieve preliminary local fixation.Samples of vaginal mucosa were collected and fixed by immersion for transmission electron microscopy,scanning electron microscopy,optical microscopic immunohistochemistry,and special staining.Results After preliminary fixation,the mucosa surface of the samples had relatively large quantities of parasites.T.vaginalis adhered like amoeba to the epithelial surface and its fingerlike pseudopod penetrated into the intercellular space of epithelia as was observed by transmission electron microscopy.Conclusion In situ fixation and fixation by immersion those samples were used to prepare specimens with large quantities of T.vaginalis,and the characteristics of the morphological structure of the parasite were closer to those observed during adhesion.
