摘要
目的探讨应用阿托伐他汀对蛛网膜下腔出血(SAA)后脑血管痉挛(CVS)的干预作用。方法取健康SD雄性大鼠30只,随机将大鼠分为假手术组、SAH组、阿托伐他汀组,每组10只大鼠。以上各组分别于第1次手术后7d灌注处死,取基底动脉行HE染色,测量其内径周长和管壁厚度;TUNEL法检测BA平滑肌细胞的凋亡率。结果假手术组、SAH组和阿托伐他汀组基底动脉的内径周长和管壁厚比较,三组之间两两比较均有统计学意义(P<0.05或P<0.01)。各组基底动脉平滑肌细胞的凋亡率比较,三组之间两两比较均有统计学意义(P<0.05)。结论阿托伐他汀可以诱导基底动脉平滑肌细胞的凋亡,早期应用对SAH后引发的CVS有一定的缓解作用。
Objective To investigate the effect of atorvastatin on cerebral vasospasm (CVS) after subarachnoid hemorrhage (SAH). Methods Thirty healthy Sprague- Dawley male rats were randomly divided into three groups: Sham- operation group(n= 10) ,SAH group (n= 10) and atorvastatin group (n= 10). All rats were sacrificed at 7th day after operation and got the basilar arter- ies(BA) for HE staining. The perimeter of inside diameter and the thickness of vessel wall were measured. The apoptosis rate of basi- lar artery vascular smooth muscle cells(VSMCs) were detected by TUNEL assay. Results The perimeter of inside diameter and the thickness of vessel wall of basilar arteries were different significantly in sham- operation group and SAH group,SAH group and ator- vastatin group,and sham- operation group and atorvastatin group(P^0.05). The apoptosis rate of the basilar artery VSMCs were statistical significance in sham- operation group and SAH group, SAH group and atorvastatin group, and sham- operation group and atorvastatin group(P%0.05). Conclusion Early applications of atorvastatin for SAH could relieve the cerebral vasospasm of the basi- lar arteries by inducing apoptosis in basilar artery VSMCs.
