In this paper, research advances on several technological puzzles, which had seriously blocked the development of plant tissue culture,had been summarized in detail,including contamination, browning or darkening, hyperhydric plantlet and genetic variation,etc.Furthermore,the author discussed the causes and resoluble methods based on the achievements in recent years in the world,combined with the experiences of whom,so as to offer useful reference for further research in this field.
