摘要
以MTT法检测了亚硒酸钠(Na2SeO3)对斑点叉尾鮰卵巢细胞(CCO)生长及抗氧化能力的影响,并在倒置显微镜下观察了CCO形态。结果表明:Na2SeO3对CCO的作用呈浓度和时间依赖性。低浓度的亚硒酸钠对细胞的形态和生长没有显著的影响,而随着亚硒酸钠浓度的增加(≥5μmol/L)和作用CCO的时间的延长(≥24h),细胞生长受到抑制,细胞形态出现损伤样变化,且随着Na2SeO3浓度的增加,抑制作用增强。高浓度Na2SeO3(≥10μmol/L)作用CCO24h,细胞内的超氧化物歧化酶(SOD),谷胱甘肽过氧化物酶(GSH-Px)活性显著下降,乳酸脱氢酶(LDH)漏出率显著增加,脂质过氧化水平显著升高,提示细胞受到严重氧化损伤。
The effects of sodium selenite(Na2SeO3) on the morphology,growth and antioxidant capacity of channel catfish ovary cell(CCO) were investigated by inverted microscope and the method of MTT.The results showed the effects of Na2SeO3on CCO were in dose response and related with time.Low doses of Na2SeO3 had little effect on CCO,while higher dose(≥5 μmol/L) significantly inhibited the cell viability and finally induced cell death.The longer the incubation time was,the stronger the damage was.Na2SeO3 at high concentration(≥10 μmol/L) induced CCO oxidative damage due to its oxidant properties.Significant decreases in activities of SOD and GSH-Px were observed.The level of lipid peroxidatioin(the content of MDA) and the release of LDH increased significantly.
