肠吉泰对内脏高敏感性模型大鼠直肠组织神经生长因子的影响

Effects of "Chang Ji Tai" on Rectal Nerve Growth Factor in Rats with Visceral Hypersensitivity

摘要 目的:研究中药制剂肠吉泰对内脏高敏感性模型大鼠直肠组织中神经生长因子(NGF)的影响。方法:40只新生SD大鼠随机分成正常组、模型组、肠吉泰低剂量组和高剂量组,每组10只。采用醋酸刺激法建立内脏高敏感性大鼠模型。肠吉泰各治疗组分别每日给予低剂量(2 g/ml)和高剂量(3.75 g/ml)中药灌胃,正常组和模型组每日分别给予等量去离子水灌胃。治疗4周后,采用直肠气囊扩张法记录引起大鼠腹壁抬起和弓背抬起的压力值以评估内脏敏感性;采用ELISA法检测直肠组织NGF含量。结果:与正常组比较,模型组和肠吉泰低剂量组引起大鼠腹壁抬起和弓背抬起的压力值均明显降低(P<0.01,P<0.05),直肠组织NGF含量明显增高(P<0.05);肠吉泰高剂量组与正常组比较,压力值和直肠组织NGF含量均无明显差异(P>0.05)。结论:肠吉泰能通过降低肠道组织NGF含量来降低内脏敏感性。 Objective:To study the effects of "Chang Ji Tai"(CJT) on nerve growth factor(NGF) in the rectum of the rats with visceral hypersensitivity.Methods:Forty neonatal Sprague-Dawley rats were randomly divided into 4 groups: normal group,model group,CJT low-dosage group and CJT high-dosage group,10 rats in each group.Visceral hypersensitivity model was induced by intrarectal acetic acid irritation.CJT-treated groups were given low-dosage(2 g/m) and high-dosage(3.75 g/m) CJT respectively every day,while normal group and model group were given deionized water.After 4-week treatments,rectal balloon distension was applied to evaluate visceral hypersensitivity and rectal NGF was detected with ELISA.Results:Compared with the normal group,the pressures inducing abdominal retraction and back hunch in model group and CJT low-dosage group were significantly lower(P0.01,P0.05),while the rectal NGF levels were significantly higher(P0.05).The pressures and NGF level in CJT high-dosage group were similar to those in normal group(P0.05).Conclusion:CJT can reduce the visceral hypersensitivity by decreasing rectal NGF level.
