珍珠菜提取物ZE4对SMMC-7721肿瘤细胞凋亡的诱导作用 被引量:6

摘要 目的探讨珍珠菜提取物(ZE4)诱导肿瘤细胞凋亡的作用。方法采用AO-EB双染法、Hoechst染色法观察ZE4给药后对人肝癌细胞株SMMC-7721肿瘤细胞形态学的影响;免疫荧光法检测ZE4给药后SMMC-7721细胞内凋亡蛋白及其配基(Fas/FasL)、肿瘤坏死因子相关凋亡诱导配体(TRAIL)、可溶性死亡受体5(DR5)、抑制凋亡蛋白(Bcl-2)及调控蛋白胱氨酸蛋白酶(Caspase3)的表达。结果Hochest染色和AO/EB双染结果表明,100μg/mlZE4可诱导SMMC-7721细胞发生凋亡,细胞凋亡的数目和形态改变随给药时间延长而变化明显;免疫荧光法检测结果表明,ZE4可通过上调Fas/FasL基因,上调TRAIL及DR5表达,下调Bcl-2表达,上调Caspase3表达等途径诱导细胞凋亡。结论珍珠菜提取物ZE4可诱导SMMC-7721肿瘤细胞发生凋亡,具有良好的抗肿瘤效果。 Objective To investigate the effects on apoptosis of cancer cells induced by ZE4,the extract from Lysimachia clethroides duby. Methods Effects of ZE4 on morphology in human cancer cells,SMMC-7721,were observed by AO/EB,Hochest. Immunofluorescence analysis was used to investigate the expression of Fas/Fas L,TRAIL,DR5,Bcl-2 and Caspase 3 in SMMC-7721 after ZE4 was added. Results The apoptosis of SMMC-7721 could be induced by 100 μg/ml ZE4. The amount and shape of the apoptosis changed obviously while the induced time changed. Through immunofluorescence analysis,it showed that ZE4 could induce the apoptosis of SMMC-7721 by increasing gene expression states of Fas/Fas L,TRAIL,DR5,Caspase 3 and decreasing gene expression states of Bcl-2. Conclusions ZE4 can obviously induce the apoptosis of SMMC-7721,indicating a potent effect on antitumor of ZE4.
