摘要
1986年,在国内尚无批量生产科研用无特定病原体动物(Specificpathogenfreeanimals)简称(SPF)和清洁动物(Cleanconventionalanimal)简称(CCV)的情况下,为尽快繁育出国际同类实验动物而开始了此项研究工作。采取的主要方法是:土洋结合,土法上马,改造原有的开放饲养动物设施,采用隔离器和洁净饲育柜,批量繁育上述实验动物。取得的主要成果是:经动管会监测基本合格,认为改造后的动物饲育区,达到了繁育CCV动物的生活环境条件。饲育柜、饲育室和走廊内的温度常年保持在23.5℃~28.0℃之间,(最高不超过29.2℃)。从饲育柜→饲育室→走廊,气压逐渐降低,形成了良好的气压梯度。柜内落下菌实验检查结果为0个/m2,噪声不超过56dB。批量繁育出的SPF和CCV实验动物包括:BALB/c/ola/ILAS/CIL;C57BL/6J/ola/ILAS/CIL;DBA/2N/ILAS/CIL;T739/CIL;Wistar/ILAS/CIL;昆明鼠(SSB)/CIL;BALB/c-nu。经主管部门监测和鉴定。
Since there was no moderate scale production for the specific pathogen free(SPF)and clean convertional animals(CCV) for the requirement of various laboratories till 1986 inour country, a research a plan was worked out for installing a production line carrying on mass production of qualified SPF and CCV animals as soon as possible. The main scheme adopted at that time was to integrate the skills at home with those abroad but starting forward by putting the reliance on our indigenous technique. We had remodeled the facilities required and fabricated the isolators as well as the clean breeding hoods instead of the equipment originally used for the open rearing in order to breed the qualified animals. The measures made the program a great progress and success, and the amimals bred had been approved by the Committee for medical experimental animals control in Beijing. In conclusion, the remodeled area and facilities were qualified for the standard of the living environment required for the breeding of clean conventional animals. The requirements were as following; (1)the temperature inside the breeding hoods, rearing rooms, and corridors should be kept constantly between23.5℃ to28.0℃, not over 29.2℃. (2) should the air pressure be kept constantly inside the breeding hood and reduced progressively in the rearing room even lower in the corridor in order to form a gradient change of the air pressure.(3)the bacteria appearance rate inside the breeding hood should be 0/m 2.(4)noise allowance was less than 56dB. The SPFand CCV animals were provided through this moderate scale production program included BALB/c/ola/ILAS/CTL, C 57 BL/6J/ola/ILAS/CIL, DBA/2N/ILAS/CIL, T739/CIL/, BALB/cnu. All these animals were approved by the medical experimental animals control department accompanied with the qualification certificates. These SPF and CCV animals had met the needs of various laboratories of Chinese Academy of Medical sciences and had been provided for 20 and even more laboratories distributing in more than 10 provinces. This was the 1st attempt carried out in this country and it seems still meaningful practically even under the current situation. \ \
