从大鼠正常及肾损伤状态下肝肾氧化应激角度探讨“肝肾同源” 被引量:5

The study of "liver and kidney sharing the same origin" in term of oxidative stress in normal and glycerol-induced renal injury rats

摘要 目的观察正常及肾损伤状态下肝肾抗氧化酶系统,以探讨肝肾抗氧化功能之间的内在联系,从氧化应激角度探讨"肝肾同源"。方法50%甘油肌内注射建立大鼠急性肾小管损伤模型,测定正常及损伤大鼠肝脏和肾脏抗氧化酶系统的活性变化。结果在急性肾损伤的不同时期(给药6h和56h),除GSH-Px外肾脏其他抗氧化酶均明显下降。与对照组相比,甘油处理6h和56h后肝脏GST、GSH-Re和SOD同样也下降,但GSH、GSH-Px和CAT则代偿性增强。相关分析表明,肝和肾CAT、GSH-Re、SOD和GST之间相关系数为-0.802(P<0.01)、0.601(P<0.05)、0.563(P<0.05)和0.667(P<0.05)。结论肾损伤时肾脏抗氧化功能降低的同时肝脏抗氧化功能也发生改变,且肝肾部分抗氧化酶存在明显的正或负相关,提示肝肾生理病理功能存在某种内在联系。 Objective To observe the relationship between renal and hepatic antioxidative functions in normal and glycerol-induced kidney injury. Methods Murine acute renal tubular injury model was established via 50% glycerol (v/v) injection. Renal histopathology,blood urea nitrogen (BUN) and creatinine (Cr) were examined. Glutathione and malondialdehyde contents,glutathione peroxidase (GSH-Px),glutathione reductase (GSH-Re),glutathione S-transferase (GST),catalase (CAT) and superoxide dismutase (SOD) activities were assessed as anti-oxidative parameters. Results Compared with control group,the activities of renal antioxidative enzymes significantly decreased except for GSH-Px. The activities of GST,GSH-Re and SOD also significantly decreased,while compensatory increase was observed in GSH,GSH-Px and CAT. The correlation coefficients of CAT,GSH-Re,SOD and GST between liver and kidney were -0.802 (P0.01),0.601 (P0.05),0.563 (P0.05) and 0.667 (P0.05),respectively.Conclusion Renal antioxidative founction is reduced in glycerol-induced kidney injury with simultaneous impairement of hepatic antioxidative function. Furthermore,there are significant negative or positive correlations some antioxidative enzymes between liver and kidney,suggesting interactions between liver and kidney,or "liver and kidney sharing the same origin".
