摘要
目的探讨不同热剂量对PLC细胞株的杀伤效应及机体免疫效应,为本地区高发肺癌临床开展热疗提供初步实验依据。方法体外实验采用MTT法检测不同热剂量对PLC杀伤率,得出适宜的杀伤热剂量,为体内试验提供依据。体内试验选取45℃与55℃作为实验组,进行肿瘤体积测量、T细胞CD4及CD8亚群检测、IL-2浓度监测。结果体外实验结果表明于44-46℃及54-56℃区间出现较强细胞杀伤,且联合DDP组呈现更强杀伤效应;体内实验结果表明45℃组与55℃组均较对照组在抑瘤效应及免疫效应具有显著差异,尤以55℃组为甚。结论热消融温度(55℃)在较短时间内杀伤肿瘤细胞,呈现出抑瘤明显且免疫效应明显等特点,值得临床参考。
OBJECTIVE:To approach PLC cell strain lethal effect and anti-tumor immunological effect by different heat dose,in order to supply experiment evidence for native lung cancer carry out thermotherapy.METHOD:Extraorgan experiment was adopt MTT to detect lethal effect by different heat dose,so that to obtain suitable heat dose in order to vivo experiment.Vivo experiment was adopt 45℃ and 55℃ experimental group,to execute measure tumor volume、detect CD4 and CD8 T-cells、monitor IL-2 concentration.RESULTS: The result of extraorgan experiment that 44~46℃ and 54~56℃ interval had the stronger lethal effect,and combine DDP was better than another experiment group.The result of vivo experiment that 45℃ group and 55℃ group have better lethal effect and anti-tumor immunological effect than control group,above all 55℃ group.CONCLUSION:The ablation temperature(55℃) could damage tumor cells in shorter time,and have stronger anti-tumor immunological effect,to be worth clinical reference.
