摘要
利用紫外—可见吸收光谱和荧光光谱,研究了5—氟尿嘧啶(5—Fu)与牛血清白蛋白(BSA)之间的相互作用.发现在pH为7.4三羟甲基胺基甲烷—盐酸缓冲溶液(Tris—HCl)中,随着5—Fu浓度的增加,BSA的特征紫外吸收峰逐渐增强,特征荧光峰的荧光强度逐渐减弱,说明它们之间存在相互作用.计算得到25℃时5—Fu与BSA的表观结合常数K_Λ为7.1×10~4L·mol^(-1),结合位点数为1.18.5—Fu与BSA的相互结合作用为荧光静态猝灭过程.
In pH 7.4 Tris-HCI buffer, the interaction between antitumor drug 5-fluroureacil (5-Fu) and bovine serum albumin (BSA) was investigated by Fluorescence and UV-Vis absorption spectroscopy. When 5-Fu added into the solution containing BSA, the fluorescence intensity of BSA decreased with the increasing of the 5-Fu concentration, while the UV absorption increased. The binding constant KA of 5-Fu with BSA was found to be 7. 1 × 10^4 L mol^-1 ,and the number of binding site is 1.18 at 25℃. The Stern -Volmer curve illustrates that the combination reaction of 5-Fu and BSA is a static quenching process.
