摘要 PCR-SSCP是一种以PCR为基础的单链构象多态性分析技术,是DNA已知突变的检测或未知变异分析中常用和实用的技术之一。影响SSCP试验效果的因素有很多,本研究主要对凝胶浓度和是否添加甘油两个因素进行分析与探讨。结果表明,凝胶浓度12%和添加甘油终浓度10%的条件下可以得到满意的结果。 Polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) is an assay according to the SSCP from PCR production on polyacrylamide gel electrophoresis (PAGE),which have been the most widely used and practical approaches for detecting known or unknown variations of DNA. There are many factors affecting the results of PCR-SSCP.In this study,analysis and discussion were carried out on influential factors,including gel's concentration and presence or absence of glycerol. The results showed that the optimal experiment conditions were as follows,gel's concentration of 12% and glycerol's concentration of 10%.
