基因表达谱芯片技术筛选颅内动脉瘤相关基因的初步研究 被引量:1

Preliminary study of screening the intracranial aneurysm related genes in peripheral blood

摘要 目的寻找动脉瘤患者外周血中基因表达谱特征并探讨其意义。方法对120例研究者(包括60例患者和60例对照)的外周血样本进行全基因表达谱分析。提取颅内动脉瘤患者的总RNA,然后应用BiostarH-140s基因芯片技术和GenePixPro3.0软件分析颅内动脉瘤患者和正常对照组的基因表达谱,发现差异表达基因,应用RT—PCR方法对差异基因进行验证。结果颅内动脉瘤患者与正常对照组比较,发现85个差异表达基因,其中表达下调的基因31个,表达上调的基因54个。RT—PCR结果证实了基因芯片技术的准确性和可靠性。结论基于患者外周血基因表达谱分析所得的差异基因,有望成为颅内动脉瘤早期诊断最有潜力的标记物。 Objective Our goal was to identify a distinct gene expression signature in peripheral blood that may identify individuals at risk for IA. Method Whole genome gene expression profiles from 120 peripheral blood samples (collected from 60 individuals with IA and 60 controls ) were analyzed. The patients with IA were collected to obtain the total RNA. The BiostarH - 140s gene chip and the GenePix Pro 3.0 software were used to analyze the gene expression map of IA patients and the normal,then the different expressed genes were found. Reverse transcription - polymerase chain reaction ( RT - PCR ) was performed on 2 genes as a confirmatory test. Results There were 85 genes differently expressed between the IA patients and the normal, there were 31 genes low expressed, and 54 genes were high expressed. RT - PCR results confirmed the differential expression fbund by the gene chip microarray. Conclusions The classifier genes identified in this study, define a set of promising potential diagnostic markers, setting the stage for a blood - based gene expression test to facilitate early detection of IA.
