The effects of the artificial synthetic E - selectin on inflammation factors and proliferation of VSMCs after carotid artery anastomosis in rats

摘要 目的探讨人工合成E-选择素对大鼠颈总动脉吻合后炎性因子和血管平滑肌细胞增殖的影响。方法将SD大鼠随机分为三组(对照组、手术组和治疗组)。对照组分离颈总动脉后股静脉注射生理盐水,手术组颈总动脉吻合后股静脉注射生理盐水,治疗组颈总动脉吻合后股静脉注射人工合成E-选择素。术后4h、1d、3d和7d用ELISA法检测血清中肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)的浓度。术后14d取吻合口血管行免疫组化染色,计算增殖细胞核抗原(PCNA)蛋白阳性表达个数和积分吸光度值(IOD)。结果人工合成E-选择素治疗组,血清中TNF—α水平在血管吻合术后4h、1d和3d时低于手术组;IL-6的水平在血管吻合术后4h、1d和3d时低于手术组;PCNA阳性细胞数和IOD也高于对照组,但低于手术组。结论人工合成E-选择素能够抑制动脉吻合后的炎性反应,减少血管平滑肌细胞增殖,减轻血管吻合口狭窄。 Objective To explore the effects of artificial synthetic E -selectin on inflammation factors and proliferation of vascular smooth muscle cells (VSMCs). Methods A total of 90 SD rats were divided randomly into 3 groups: the control group, the surgery group and the therapy group. The right carotid arteries were anastomosed in the surgery group and in the therapy group. Artificial synthetic E - selectin was used in therapy group. The serum levels of tumor necrosis factor -α and interleukin -6 were detected with ELISA assay at 4 h, 1 d, 3 d and 7 d after the operation. After 2 weeks, the vessels were stained by immunity histochemistry method, then calculated the counts and integrated optical density (IOD) value of protein expression of proliferation cell nuclear antigen (PCNA) with microscope. Results The serum levels of tumor necrosis factor - α and interleukin -6 in the therapy group are lower than the surgery group at 4 h, 1 d, 3 d and 7 d after the operation. The PCNA positive cell counts and IOD in the therapy group were lower than the surgery group. Conclusion Artificial synthetic E -selectin can inhibit the inflammation reaction, decrease proliferation of VSMCs and lighten the stenosis after carotid artery anastomosis in rats.
