摘要
目的观察传统抗痨方剂百合固金汤含药血清对巨噬细胞表面模式识别受体的影响,以探讨传统医药抗结核的可能作用机制。方法按常规方法制备百合固金汤含药兔血清。以含药血清作用U937单核巨噬细胞24 h后,分别采用RT-PCR法和流式细胞仪检测巨噬细胞表面CR1、CR3、CD14、CD43、TLR2及TLR4的表达。同时设空白兔血清为对照。结果 RT-PCR和流式细胞仪结果均显示百合固金汤含药血清可使巨噬细胞表面模式识别受体CR1、CD14、TLR2、TLR4表达显著增高(P<0.05);而CR3仅在蛋白表达水平显示差异(P<0.05);CD43表达则未见显著差异(P>0.05)。结论传统抗痨方剂百合固金汤的抗结核作用可能通过对巨噬细胞表面模式识别受体的调节而发挥作用。
To investigate the effects of Bai He Gu Jin Tang Drug-serum on expression of pattern recognition receptor (PRR) in macrophages for evaluating the possible mechanism of Bai He Gu Jin Tang Drug against tuberculosis. We prepared the Bai He Gu Jin Tang Drug-serum and blank serum. The two serum samples were used to incubate with U937 monocyte-derived macrophages for 24 hours. Then, RT-PCR and FACS were used to detect the mRNA expression of CR1, CR3, CDI4, CD43, TLR2 and TLR4 in macrophages. Compared to blank serum group, the Bai He Gu Jin Tang drug-serum group significantly increased the mRNA and protein expressions of CR1, CD14, TLR2 and TLR4 in macrophages (P 〈 0.05), and up-regulated the CR3 protein expression (P 〈 0.05) while its mRNA expression didn't change. But the expression of CD43 protein and mRNA were no significant differences between the two goups (P〉0.05). All the results come to the conclusion that Bai He Gu Jin Tang maybe treat the tuberculosis by regulating these PRRs in macrophages.
