摘要 目的:研究人骨肉瘤细胞株MG63上功能性N-甲基-D-天冬氨酸(NMDA)受体的表达。方法:利用钙离子成像系统检测NMDA或谷氨酸对MG63细胞内游离钙离子浓度([Ca2+]i)的影响。结果:仅6.4%MG63细胞对NMDA产生[Ca2+]i升高反应,对谷氨酸则没有反应。多数MG63细胞呈现自发性的钙离子震荡现象,且该现象不能被MK801所阻断。结论:MG63细胞表达的功能性NMDA受体很少。 Objective To investigate the expression of functional NMDA (N-methyl-D-aspartic acid) receptor in human osteosarcoma cell line MG63. Methods Calcium ion imaging technique was used to observe the effects of NMDA and glutamate on the level of [Ca^2+]i in MG63 cells. Results Only 6.4 percent of MG63 cells showed [Ca^2+]i elevation in response to NMDA, and no response was observed with glutamate. Most of the MG63 cells showed the phenomenon of spontaneous calcium oscillation, which could not be blocked by NMDA receptor antagonist MK801. Conclusions These results indicates that only small amount of functional NMDA receptor is expressed in MG63 cell line.
