中肺合剂对Lewis肺癌小鼠肿瘤组织p53和p21表达的影响 被引量:3

Effect of Zhongfei Mixture on p53 and p21 Expression in Mice Bearing Lewis Lung Cancer Tissues

摘要 [目的]观察中肺合剂对小鼠Lewis肺癌肿瘤组织p53、p21基因表达的影响。[方法]C57BL/6荷Lewis肺癌小鼠50只,随机分生理盐水对照组,中肺合剂低、中、高剂量组,环磷酰胺组,计算抑瘤率。EnVision免疫组化方法测定中肺合剂对小鼠Lewis肺癌组织p53、p21表达的影响。[结果]中肺合剂低、中、高剂量组和CTX组对移植性荷Lewis肺癌小鼠抑瘤率分别为20.57%、41.18%、24.80%和55.42%,以中剂量组抑瘤效果最好,与生理盐水组相比,有非常显著性差异(P<0.01)。中肺合剂能使Lewis肺癌荷瘤小鼠肿瘤组织p53表达呈降低趋势,p21表达呈增强趋势,但与生理盐水组比较,无显著性差异(P>0.05)。[结论]中肺合剂可能不是通过p53来发挥其抑瘤作用。 [ Purpose] To investigate the effect of Zhongfei Mixture(ZM) on the expression of p53 and p21 in mice bearing Lewis lung cancer tissues. [Methods] Fifty C57BL/6 mice bearing Lewis lung carcinoma were divided into 5 groups: saline(NS) group, the low-moderate-and high-dose with ZM groups and Cyclophosphamide(CTX) group. The expressions of p53 and p21 in tumor tissues were detected normal by EnVision two-step immuno-histochemistry staining. [Results] The inhibitory rate of the low-, moderate-, and high-dose with ZM groups and of CTX group was 20.57%, 41.18%, 24.80%, and 55.42% respectively. The best inhibitory potential was found in moderate-dose with ZM group, with significant difference(P〈0.01) verus NS group. ZM was able to decrease p53 expression and increase p21 expression in the mice bearing Lewis lung cancer tissues, but its effect showed no significant difference compared with NS control group (P〉0.05). [Conclusions] The anti-tumor Effect of ZM may not be carried out by p53.
