摘要
目的探讨前列腺凋亡反应因子(prostate apoptosis response-4,Par-4)和人端粒酶逆转录酶(human telome rasereverse transcriptase,hTERT)两种蛋白在靶向hTERTmRNA干扰前后的喉癌中的表达及其相关性。方法基于RNAi技术原理,用质粒pEGFPshTERT干扰体外培养的喉癌Hep-2细胞,免疫细胞化学分别检测干扰前、后12、24、36、48小时的喉癌细胞中hTERT和Par-4的表达情况;用质粒pEGFP-shTERT治疗喉癌裸鼠模型,量子点超敏荧光检测治疗前,治疗后6、10、14天的喉癌组织中hTERT和Par-4的表达情况。结果用质粒pEGFP-shTERT干扰前喉癌中hTERT呈高表达,Par-4仅在胞浆表达,干扰后hTERT表达降低,Par-4胞核表达增多,各组间的蛋白表达差异有统计学意义;相关性分析显示:RNAi干扰前后,喉癌中Par-4和hTERT的表达成负相关(P<0.05,r=-0.908)。结论在喉癌中,靶向敲除hTERT mRNA后,伴随hTERT表达降低和细胞凋亡的发生,Par-4表达逐渐增多,提示Par-4与hTERT相互作用可能对hTERT的非端粒酶依赖的抗凋亡途径有影响。
OBJECTIVE To study the expression of prostate apoptosis response(Par-4)and human telomerase reverse transcriptase(hTERT)and their relationship in laryngeal cancer. METHODS RNAi technique was used in the experiment. The expression of Par-4 and hTERT was detected by quantum dots(QDs)-based immunofluoresense method in xenograft laryngocarcinoma from nude mice and immunocytochemistry in laryngocarcinoma cells Hep-2. RESULTS After RNAi with laryngocarcinoma, the expression of hTERT was inhibited and the expression of Par-4 was increased. The difference among groups was statistically significant(P0.05). There was a negative correlation between Par-4 and hTERT(P0.05,r=-0.908). CONCLUSION The increased expression of Par-4 accompanying with the inhibition of hTERT indicates that Par-4 interacting with hTERT may have some effect on telomerase-independent anti-apoptosis of hTERT.
