替米考星口服后在肉鸭体内的生物利用度研究

摘要 目的为了解替米考星在肉鸭体内的药代动力学特性及生物利用度,为替米考星在临床应用作出正确评价与指导。方法8只健康成年北京肉鸭,采用静脉注射和口服两种途径分别以替米考星5mg/kg和20mg/kg体重剂量给药,进行体内药代动力学研究,计算其生物利用度。动物给药后72h内分点采血,用反相高效液相色谱法测定不同时间点血浆中的药物浓度。采用3p97药代动力学软件处理数据,替米考星两种给药途径的药时数据均符合二室开放模型。结果静脉注射给药(5mg/kg体重)的主要药代动力学参数:t1/2α为0.076±0.0098h、t1/2β为7.31±0.63h、AUC为4.50±0.30/μg·ml^-1·h^-1、CL(s)为1.12±0.069/L·kg^-1·h^-1;口服给药(20mg/kg体重)的主要药代动力学参数:t1/2α为1.76±0.49h、t1/2β为25.70±4.74h、t1/2Ka为0.090±0.0092h、AUC为14.03±2.54/μg·ml^-1·h^-1、CL(s)为1.46±0.22/L·kg^-1·h^-1、tmax为0.47±0.035h、Cmax为0.75±0.056μg/ml、F为78.47%±0.15%。结论通过剂量较正法计算出口服给药后在肉鸭体内的生物利用度为78.47%±0.1483%,表明替米考星口服给药在肉鸭体内吸收良好。
