摘要
为了解析家蚕对常用农药氰戊菊酯和杀虫双的解毒机制,分别给家蚕5龄幼虫添食2种农药,观察家蚕对农药的中毒症状,采用双向电泳(2-DE)技术对添食农药的家蚕幼虫的血液蛋白进行分离,利用电喷雾电离(ESI)质谱对具有2次以上重复的显著差异蛋白点进行鉴定,并用生物信息学方法进行分析。家蚕5龄幼虫分别添食1/4LD50剂量浓度的2种农药后,均在3~12h内出现中毒症状,且血液蛋白的表达也发生了变化:在氰戊菊酯诱导下,幼虫血液中增加了纤维样蛋白、果糖-1,6-二磷酸醛缩酶、半胱氨酸蛋白酶抑制剂;在杀虫双诱导下,幼虫血液中增加了丝氨酸蛋白酶抑制剂-13;经2种农药分别诱导处理后3h,幼虫血液中都减少了体液低分子蛋白-Ⅲ的表达。推测家蚕对氰戊菊酯和杀虫双具有一定的免疫反应。
In order to investigate the detoxification mechanisms of silkworm (Bombyx mori) against Fenvalerate and Dimehypo, two pesticides in common use, silkworm larvae of the 5th instar were administered with these two pesticides to observe their poisoning symptoms. Two dimensional electrophoresis was employed to isolate haemolymph proteins from the poisoned larvae. The protein spots with significant difference which recurred over two times were identified through electrospray ionization mass spectrometry and were subsequently analyzed using bioinformatics approach. It was found that both silkworm larvae of the 5th instar showed poisoning symptoms within 3 to 12 h after being administered with the two pesticides at a dosage of 1/4 LD50. Moreover, expression of haemolymph proteins was changed. fibrillin-like protein, fructose-1,6-bisphosphate aldolase and Bombyx cysteine proteinase inhibitor were induced by Fenvalerate and serpin-13 by Dimehypo in haemolymph of the poisoned larvae. On the contrary, the expression of apolipophorin-Ⅲ in haemolymph of the silkworm larvae was reduced 3 h after the induction by both pesticides. We speculate that silkworm larvae have certain immune response to Fenvalerate and Dimehypo .
