摘要
目的参照中国药典(2005版)测定沙枣花(Elaeagnus angustifoliablossoms,EAB)中总黄酮和芦丁的含量,并初步探讨沙枣花醇提物(the alcohol extract of Elaeagnus angustifoliablossoms,AE-EAB)对四氯化碳(CCl4)诱导的小鼠急性肝损伤的保护作用。方法参考《中国药典》通过分光光度法和高效液相色谱法测定EAB中总黄酮和芦丁的含量,并通过测定小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)的活性和肝组织匀浆中丙二醛(MDA)的含量和超氧化物歧化酶(SOD)、谷胱甘肽(GSH)的活性及观察肝脏形态学改变,研究不同剂量AE-EAB对小鼠CCl4急性肝损伤模型的保护作用。结果测得EAB中总黄酮的含量为27 mg/g,芦丁的含量为0.029 mg/g。与CCl4诱导的肝损伤模型比较,AE-EAB给药组小鼠血清中ALT,AST活性明显降低(P<0.05),同时小鼠肝组织中MDA含量明显下降(P<0.01),GSH含量有所升高(P<0.05),SOD活力明显增强(P<0.01)。形态学观察,AE-EAB能明显减轻CCl4对肝组织的损伤。结论EAB含有黄酮类化合物,AE-EAB对CCl4诱导的小鼠急性肝损伤具有保护作用。
Objective To determine the contents of total flavonoids and rutin in Elaeagnus angustifolia blossoms(EAB) according to Technical Standard For Chinese Pharmacopoeia (2005 version) , and to study the protective effects of the alcohol extract of Elaeagnus angustifolia blossoms( AE - EAB) on acute hepatic injury induced by carbon tetrachloride (CCl4 ) in mice. Methods Spec- trophotomctrie method and high performance liquid chromatography were used to determine the contents of total flavonoids and rutin in EAB, according to Technical Standard For Chinese Pharmacopoeia (2005 version). The levels of ALT, AST in serum and MDA , SOD and GSH in liver were measured and the hepatic histological changes were observed by optical microscope in order to study the protective effects of different doses of alcohol extract of AE - EAB on acute hepatic injury induced by 0.1% CCl4 in mice. Results The content of total flavonoids of EAB is 27mg/g ( 27mg total flavonoids in lg EAB ) , and that of rutin was 0.029mg/g(0. 029 mg rutin in lg EAB). Compared with model group, the levels of ALT, AST in serum were obviously decreased ( P 〈 0.05 ), and the level of MDA in liver was also lowered ( P 〈 0.05 ), but the activities of GSH and SOD in liver were markedly improved (P 〈 0.05, P 〈 0.01 ) after pretreatment with AE - EAB. AE - EAB ameliorated the hepatic pathological chan- ges too. Condusion The results show that EAB contains flavonoids, and AE - EAB has protective effects on acute hepatic injury induced by CCl4 in mice.
