幼鼠内毒素血症脾内单核-巨噬细胞等动态分布

Dynamic Distribution of Mono-Macrophages and Neutrophilic Granulocytes in Spleen of Young Rat with Endotoxaemia

摘要 用组织化学方法和生化方法探讨了幼龄大鼠内毒素血症脾急性非特异性免疫反应。腹腔注入内毒素(i.P.LPS)1h,脾内碱性磷酸酶(ALPase)阳性细胞开始增多,至12h红髓和边缘区内呈现大量的ALPase阳性细胞,24~72h这些阳性细胞逐渐减少至正常。电镜下证实这些阳性细胞多数为单核-巨噬细胞和中性粒细胞。腹注LPS6~12h,脾索和淋巴小结周围呈现较多的一氧化氮合成酶(NOS)阳性细胞。腹注LPS6~12h生化方法测定牌匀浆NOS和NO的水平显示两者均升高。本研究的结论是:(1)LPS可刺激脾内单核-巨噬细胞和中性粒细胞增多,表明它们有明显的动态分布;(2)脾脏NOS活性反应和NOS及NO含量有严格的时间性;(3)就幼龄大鼠来说,腹注LPS其有效毒性只能维持在24h之内。 Acute nonspecific immunoresponse of spleen in young rat with endotoxaemia was approached by using histochemical method and biochemical techniques. Intrapritoneal injection of LPS into the young rats resulted in an increase of ALPase-positive cells in spleen at 1 h after LPS injection. A nnmber of ALPase-postitve cells located in the red pulp and marginal zone at 12 h. These positive cells gradually reduced to normal distribution from 24 to 72 h after us injection. Most of the positive cells were verified as meno-macrophages and neutrophilic granulocytes electron microscopy. There were more NOS postitve cells in spleen cord and around lymphoid follicle from 6 to 12 h after LPS injection. An elevation of NOS and NO levels in spleen homegenate was tested by using biochemical techniques from 6 tu 12 h after LPS injection. The conclusions are as followe: (1) As mono-macrophages and neutrophilic granulocytes increase in number by stimulation of LPS, it is suggested that they present a pronounced dynamic distribution. (2)There is strict timeliness of NOS, active reaction and NOS, NO increase in spleen.(3 )Powerful toxic action only persists in 24 h in young rats after us injection.
