鸡传染性贫血病毒与禽网状内皮增生症病毒二重PCR检测方法的建立

Establishment of a Double PCR Assay for Detection of Chicken Anaemia Virus and Reticuloendotheliosis Virus

当前养鸡业中鸡传染性贫血病毒、禽网状内皮增生症病毒混合感染的发生率日益增高,为提高CIAV、REV检测效率,根据GenBank上登录的CIAV全基因序列和REV的LTR序列分别设计了2对引物,建立了REV、CIAV二重PCR检测方法,另外还比较了2种不同的DNA提取方法。结果显示,该方法扩增目的条带清晰、敏感性高、特异性好。应用建立的方法对临床7份病料样品检测,检出5份CIAV、REV阳性,表明建立的二重PCR方法能有效用于CIAV、REV混合感染的临床诊断。

The co-infection incidence of chicken anaemia virus（CIAV）,reticuloendotheliosis virus（REV） are increasing in present poultry industry,in order to enhance the detection efficiency of CIAV and REV,according to the sequence of whole gene of CIAV and the LTR of REV published in GenBank,two pairs of primers were designed,the double PCR of REV,CIAV was established.Moreover,two kinds of extraction method of DNA were compared.The results showed that the object fragments were amplified and hold high sensitivity and good specificity.Results showed the establishment of double PCR assay was effective in the clinical diagnosis of co-infection of CIAV and REV.
