细胞凋亡基因ICE在抗癌药处理后的食管癌细胞中的表达

摘要 应用顺铂诱导体外培养的人食管癌细胞发生凋亡,在光镜下观察细胞的形态学改变,用免疫细胞化学方法染色以检测肿瘤细胞白介素1β转化酶(ICE)基因的表达情况。结果显示,顺铂处理后的肿瘤表现为出芽、与相邻细胞分离、核碎裂、凋亡小体形成及其被吞噬现象。ICE蛋白在未用药处理的对照细胞呈阴性表达,而发生凋亡的细胞及凋亡小体呈阳性表达,呈阳性表达的细胞数目及着色强度随药物作用时间延长而增加,然而一些不具备凋亡形态特征或处于凋亡前状态的肿瘤细胞ICE蛋白也呈阳性表达。结果表明ICE基因可能参与了顺铂诱导肿瘤细胞凋亡的调节。 Cisplatin was used to induce apoptosis in human esophageal carcinoma cells cultured in vitro.The morphological changes were observed under light microscope.The expression of interleukin-1β-converting enzyme(ICE) in the tumor cells was examined by immunocytochemical staining method.The results showed that apoptotic cells exhibited budding,detaching from their neighbours, karyorrhexis, the formation of apoptotic bodies and the phenomena of their being phagocytosed.ICE were expressed negatively in control cells while expressed posively in apoptotic cells and apoptotic bodies. The number and the staining intensity of positive cells increased with the extension of exposure to the drug.However,some cells without morphological changes of apoptosis or in a preapoptotic state expressed ICE protein positively.The results indicate that ICE gene may be involved in the regulation of cisplatin-induced apoptosis in the tumor cells and ICE protein may play a certain role at an early stage of apoptosis of tumor cells induced by cisplatin.
