摘要 目的使用同种兔骨髓间充质干细胞(BMSCs)修复关节软骨缺损。方法体外培养扩增兔BMSCs。采用软骨诱导分化培养基对P2代细胞进行成软骨诱导,同时制备兔关节软骨缺损模型。诱导后细胞与事先制备好的"壳聚糖-胶原蛋白"支架复合,移植到软骨缺损区。术后于2、4、8、16周进行组织学评分并杀死动物行组织学染色。结果术后16周移植的细胞一致分化为软骨细胞,大部分软骨缺损区被新生软骨修复。结论应用BMSCs结合"壳聚糖-胶原蛋白"复合物可以修复关节软骨缺损。 Objective Using the same BMSCs to repair articular cartilage defects. Methods BMSCs were cultured and proliferated. P2 cells were differentiated in chondrogenic medium. The induced cells then were mount in the chitosan-collagen scaffold and transplanted into the rabbits' cartilage defect model. 2, 4, 8, 16 weeks later the rabbits were killed and then histological staining was done. Results After 16 weeks most of the defects of cartilage were repaired completely. Conclusion BMSCs combined with chitosan-collagen can repair articular cartilage defects.
